چکیده

Data from many laboratory and clinical investigations indicate that CD34+ cells comprise approximately 1% of human bone marrow (BM) mononuclear cells, including the progenitor cells of all the lymphohematopoietic lineages and lymphohematopoietic stem cells (stem cells). Because stem cells are an important but rare cell type in the CD34+ cell population, investigators have subdivided the CD34+ cell population to further enrich stem cells. The CD34+/CD38cell subset comprises less than 10% of human CD34+ adult BM cells (equivalent t o <0.1% of marrow mononuclear cells), lacks lineage (/in) antigens, contains cells with in vitro replating capacity, and is predicted to be highly enriched for stem cells. The present investigation tested whether the CD34+/CD38subset of adult human marrow generates human hematopoiesis after transfer t o preimmune fetal sheep. CD34+/ CD38cells purified from marrow using immunomagnetic

Bone marrow stromal cells and their application in neural injuries

Background: This article reviews experimental and clinical studies in which neural injuries repaired with bone marrow stromal cells. History: Bone marrow contains two kinds of stem cells: hematopoietic and nonhematopoietic (stromal) stem cell. In vitro studies indicate that bone marrow stromal cells have the capacity of differentiation into other cells (such as neural cell) under treatment wit...

EXPANSION OF HUMAN CORD BLOOD PRIMITIVE PROGENITORS IN SERUM-FREE MEDIA USING HUMAN BONE MARROW MESENCHYMAL STEM CELLS

Ex vivo expansion of human umbilical cord blood cells (HUCBC) is explored by several investigators to enhance the repopulating potential of HUCBC. The proliferation and expansion of human hematopoietic stem cells (HSC) in ex vivo culture was examined with the goal of generating a suitable clinical protocol for expanding HSC for patient transplantation. Using primary human mesenchymal stem ...

Equine Bone Marrow Derived Mesenchymal Stem Cells: Isolation and Multilineage Differentiation

Objective- To evaluate growth characteristics and differentiation capacity of equine mesenchymal stem cell (eMSCs) derived from bone marrow (BM). Study design- In vitro experimental study. Animals- Four young adult horses (2-5 years old) Procedure- Cell morphology and growth characteristics of eMSCs harvested from BM were evaluated in standard culture conditions. eMSCs in passage 3 were subj...

Efficient expression of functional human MDR1 gene in murine bone marrow after retroviral transduction of purified hematopoietic stem cells.

A procedure for efficient transfer of the human MDR1 (multi-drug resistance) gene into murine hematopoietic stem cells was developed. Cells expressing Sca-1 but no lineage-specific or major histocompatibility complex (MHC) class II antigens (Lin-MHC II-Sca-1+) were enriched from 5-fluorouracil-pretreated bone marrow by Ficoll density-gradient and immunomagnetic sorting. Purified cells were cocu...
